 ificial intelligence is rapidly evolving from a passive analytical tool in
 to an active partner in scientific discovery. In the material world\, this
  transition requires systems that do more than recognize patterns: they mu
 st reason across scales\, formulate hypotheses\, design and execute invest
 igations\, learn from failure\, and revise their own representations in re
 sponse to evidence. This talk presents emerging approaches toward “super
 intelligent” discovery engines that integrate reinforcement learning\, g
 raph-based reasoning\, physics-informed neural architectures\, generative 
 models\, and autonomous tools for simulation\, experimentation\, design\, 
 and manufacturing. Particular emphasis will be placed on multi-agent syste
 ms inspired by collective intelligence in nature\, in which specialized ag
 ents collaborate\, critique one another\, and continuously improve their s
 trategies as they solve complex scientific problems. Case studies from mat
 erials science\, engineering\, and biology will illustrate how these syste
 ms can uncover hidden structure-property relationships\, generate and eval
 uate novel material and molecular designs\, connect knowledge across disci
 plinary boundaries\, and transform scientific failures into new hypotheses
  and executable knowledge. These advances point toward a new form of artif
 icial intelligence that actively expands the boundaries of human knowledge
 .\n\nShort Bio Sketch: Markus J. Buehler is the McAfee Professor of Engine
 ering at MIT\, a pioneer in AI-driven knowledge discovery and materials sc
 ience. He created graph-aware\, multi-agent AI platforms that turn heterog
 eneous data into science-grounded\, actionable insight\, powering breakthr
 oughs in materials science\, biology\, and healthcare. Buehler is among th
 e world's most-cited materials scientists and the recipient of numerous ho
 nors\, including the Feynman Prize\, ASME Drucker Medal\, J. R. Rice Medal
 \, and the Washington Award. He is a member of the U.S. National Academy o

DESCRIPTION:Autonomous robots are increasingly expected to operate in dyna
 mic\, human-centered environments. To do so safely and efficiently\, they 
 must reason about how people move and interact. In domains like driving\, 
 social navigation\, and aviation\, a common approach is to learn models of
  human motion directly from recorded data and use the resulting priors to 
 inform downstream systems like simulators and planning stacks.\n\nDespite 
 the growing availability of datasets\, benchmarks\, and modeling technique
 s\, state-of-the-art methods remain unreliable for real-world deployment\,
  often generalizing poorly to novel environments and rare events. Much of 
 this stems from recorded datasets covering few environments and few safety
 -relevant events relative to what a deployed system will ultimately encoun
 ter. Broadly\, the field has addressed this in four main ways: validating 
 autonomy stacks on the road\, collecting more data\, synthesizing relevant
  scenarios\, and adapting at test time. These are all valuable and necessa
 ry strategies\, but each is bounded by risk\, cost\, the sim-to-real gap\,
  or the difficulty of reliably detecting and adapting to a shift\, respect
 ively.\n\nThese strategies share the premise that the data we hold is insu
 fficient. This dissertation argues that such data is also underexploited a
 nd thus pursues a complementary direction\, asking how much actionable sig
 nal existing datasets already contain but current practice overlooks. We d
 o so through a recurring paradigm we call scenario characterization: descr
 ibing a scenario in terms of a property of interest and acting on that des
 cription downstream. We use this paradigm in three ways. The first is for 
 guidance and abstraction\, shaping what a model trains or optimizes. The s
 econd is for mining and benchmarking\, determining what a model trains on 
 and what is withheld to test it. The third is for analysis\, fixing the ba
 sis on which results are studied. We apply these uses across two settings:
  in-distribution generalization\, which draws mainly on guidance and abstr
 action\, and generalization under distribution shift\, which draws on all 
 three and is where the main contributions concentrate.\n\nWe further argue
  that evidence for generalization is typically gathered only within a sing
 le domain\, so a claim that holds there is rarely challenged elsewhere. Th
 is is largely because\, outside autonomous driving\, no motion domain offe
 rs comparable infrastructure and scale. This dissertation therefore contri
 butes aviation as a testbed\, introducing a large-scale framework and data
 set for airport surface movement forecasting. \n\nThrough this framework a
 nd our experimental settings\, we expose generalization failures that woul
 d otherwise have remained hidden in aggregate metrics. We also enable read
 ing aviation and driving on a common basis\, hinting at what transfers and
  what does not. Finally\, we also provide early evidence that the signal r
 ecovered through our framework may help systems gain the risk awareness ne

DESCRIPTION:Distribution shift\, i.e. a change in the distribution of inpu
 t/output data between training and test time\, remains an enduring challen
 ge in modern ML. In practice\, methods to improve robustness of ML models 
 to distribution shift are frequently heuristic and require considerable si
 de information about the shifted (target) test distribution to be effectiv
 e. In theory\, fundamental questions like why ML models are initially brit
 tle to distribution shifts\, how to best mitigate this brittleness\, and h
 ow much target data is required for this mitigation remain open.\n\nIn thi
 s talk\, we put distributional robustness under the microscope by studying
  the “group robustness” setting - where data is comprised of majority 
 and minority groups whose relative proportions shift between training and 
 test time. We present two case studies:\n\n - On the empirical side\, we s
 how that simply retraining the last layer of state-of-the-art neural netwo
 rks on a class-balanced held-out set is surprisingly effective - even if t
 he held-out set remains sizably group-imbalanced - on well-established gro
 up robustness benchmarks. We also present a data-driven\, active learning-
 inspired methodology to select and upweight minority group examples which 
 achieves comparable group robustness on these same benchmarks to state-of-
 the-art methods that do require group labels. \n - On the theoretical side
 \, we provide the first end-to-end analysis showing that shallow neural ne
 tworks learn a simpler spurious correlation and suppress the true (core) p
 redictor. We consider the core predictor to be the nonlinear XOR function 
 while the spurious correlation is linear. Our analysis and techniques reve
 al a striking phase transition between the situations of “extreme” cor
 relation and “moderate” correlation\, and could explain the success of
  existing robustness-advancing methods as well as inspire new ones.\nThis

DESCRIPTION:We devise multiview tomography from space. This talk will show
  the First Light of CloudCT\, the very first image taken since we launched
  our first satellite. This opens opportunities for scientific observations
  and technologies\,  can transform climate research and potentially even a
 ffect medical X-ray CT.  Tasks and solutions involve both novel imaging ha
 rdware and computational algorithms\, based on machine learning and differ
 ential rendering. The key idea is that advanced computing enable computed 
 tomography of volumetric scenes\, based scattered radiation.  CloudCT\, fu
 nded by the ERC\, involves 10 nano-satellites to fly in an unprecedented f
 ormation\, to capture the same scene (cloud fields) from multiple views si
 multaneously. We encounter challenges of  polarimetric self-calibration in
  orbit\, and estimation of 3D volumetric distribution of microphysical pro

DESCRIPTION:Chronic\, non-healing wounds affect 2% of the U.S. population\
 , with impaired vascular reperfusion contributing to defective closure\, p
 articularly in aging and diabetic individuals. Yet\, the cellular and mole
 cular mechanisms underlying endothelial responses during vascular regenera
 tion remain poorly defined. Zebrafish uniquely combine conserved regenerat
 ive mechanisms with extensive transgenic resources and long-term live imag
 ing capabilities\, enabling dynamic analysis of repair in vivo. We establi
 shed an adult zebrafish cutaneous wound model that pairs rotary tool injur
 ies with cellular-resolution longitudinal imaging to define vascular repai
 r dynamics. Live imaging revealed a temporal sequence of wound healing\, w
 ith neutrophil recruitment initiating within hours and peaking by one day\
 , macrophage activity and vessel regrowth increasing between 1-4 days\, an
 d vascular remodeling continuing over months. Aging disrupts this response
 \, with aged fish exhibiting accelerated neutrophil recruitment but delaye
 d vascular regrowth.\n\nTo define the molecular programs underlying endoth
 elial responses during repair\, we utilized AngioTag transgenic fish for c
 ell-type-specific TRAP-RNAseq profiling of the endothelial translatome acr
 oss wound healing stages. Profiling revealed distinct temporal phases of v
 ascular repair\, including early inflammatory activation (0-2dpi)\, angiog
 enic migration and proliferation programs (2-4dpi)\, vascular remodeling a
 nd extracellular matrix interactions (4-7dpi)\, and matrix maturation and 
 resolution-associated signatures (7-15dpi). Defining these transitions pro
 vides a framework for linking dynamic endothelial behaviors observed by li
 ve imaging to the underlying molecular programs that coordinate vascular r
 egeneration. Together\, these approaches establish a temporal atlas of end
 othelial regenerative states\, revealing how endothelial responses are dyn
 amically regulated during regeneration. This framework provides insight in
 to the molecular basis of impaired vascular repair with age and highights 
 potential new avenues for therapeutic interventions.\n\nFaculty Host: Nate

  inhibition of TMEM16A by fenamate NSAIDs\nAbstract: Transmembrane protein
  16 (TMEM16A) is a Ca2+-activated Cl- channel which enables the movement o
 f Cl- across cell membranes. TMEM16A is found in a variety of cell types\,
  including airway epithelia and vascular smooth muscle cells (VSMCs)\, whe
 re it functions in mucosal secretion or cell contraction\, depending on ce
 ll type and location. In VSMCs\, the efflux of Cl- through TMEM16A channel
 s leads to the contraction of arteries. TMEM16A has recently emerged as a 
 molecular target of nonsteroidal anti-inflammatory drugs (NSAIDs)\, in the
  cardiovascular system. NSAIDs are known to induce an increase in blood pr
 essure through COX enzyme inhibition\, which can lead to secondary effects
  including heart attack or stroke. However\, the full picture of how NSAID
 s influence vascular health remains incomplete. By studying NSAID regulati
 on of TMEM16A\, we can better understand their cardiovascular effects. I a
 m focusing on the fenamate subclass of NSAIDs\, which share a parent struc
 ture and differ in their substituents. Here I examine niflumic acid (NFA)\
 , a broadly studied fenamate with contested mechanisms of TMEM16A inhibiti
 on. To date\, I have collected data using excised inside-out patch-clampin
 g on Xenopus laevis oocytes\, a method enabling COX-independent investigat
 ion of NSAID regulation. I found that NFA inhibits TMEM16A with dose-depen
 dency and exhibits characteristics of a pore-blocking drug. I also want to
  define the structural components that drive fenamate inhibition of TMEM16
 A and affect its potency. I found that another fenamate\, flufenamic acid 
 (FFA)\, inhibits TMEM16A like NFA at equivalent concentrations\, but shift
 s the reversal potential by ~10 mV at 100 µM despite structural similarit
 y with NFA. Further characterization of these mechanistic and structural f
 eatures will provide physiologically relevant insights into how fenamate N
 SAIDs regulate TMEM16A and contribute to cardiovascular risks.\n\nJulia Ro
 se\nTitle: Shaping a chaperone: the co-occurrence of PrsA2 and the pore fo
 rming toxin LLO \nAbstract: Listeria monocytogenes (Lm) is a Gram-positive
  bacterium found within the environment as a saprophyte but maintains the 
 ability to become a facultative intracellular pathogen\, deadly to vulnera
 ble populations. Infection occurs via secreted virulence factors which all
 ows the bacterium to invade host tissues. Lm encodes a secreted chaperone\
 , PrsA2 that is critical for the activity of secreted virulence factors in
 cluding the cholesterol-dependent pore-forming toxin\, listeriolysin O (LL
 O). PrsA2 has been shown to directly associate with and protect LLO during
  infection conditions\, and PrsA2 is required for the full functionality o
 f LLO. While a relationship between PrsA2 and LLO has been established\, i
 t remains to be seen how PrsA2 clients and environmental conditions shape 
 the chaperone. To answer this question\, we explored the evolution of both
  PrsA2 and LLO in annotated strains\, to determine whether genetic diversi
 ty can be used to gain insight into the molecular mechanisms used between 
 PrsA2 and LLO. We generated bioinformatic data separating PrsA2 and LLO in
 to distinct clades based on their sequences and determined the percentage 
 of interaction between each PrsA2 and LLO clade. To confirm these percenta
 ges and visualize how evolution made this interaction more favorable\, we 
 used biophysical experiments and structural analysis. We hypothesize that 
 by relying on the genetic diversity of both proteins\, we can further eluc
 idate the mechanism by which PrsA2 interacts with LLO and have a deeper un
 derstanding on if the chaperone is shaping the client or if the client is 
 shaping the chaperone.

 ve work at Los Alamos National Laboratory. The first uses Bayesian modelin
 g to characterize source temperature and emission profiles from inertial-c
 onfinement-fusion diagnostics. The second develops a statistical pipeline 
 to turn measurements from a compact sensor flown on the International Spac
 e Station into meaningful ionospheric signal. The third builds statistical
  emulators to learn about the universe from computationally expensive cosm
 ological simulations. Across the examples\, I will focus on the interplay 
 between scientific objectives\, statistical modeling\, uncertainty\, and c

DESCRIPTION:Digital media creators\, including video editors\, live stream
 ers\, and game developers\, rely on diverse creativity tools such as game 
 engines\, video editing software\, and live streaming platforms to produce
  their work. These ecosystems are powerful\; however they often impose con
 straints on their users\, leading to rigid interfaces that fail to adapt t
 o how creators want to work. Existing tools can limit a creator’s abilit
 y to realize ambitious ideas\, offer inadequate mechanisms for communicati
 on between creators and their audiences\, and provide interfaces that do n
 ot align with how creators naturally express their preferences. The first 
 half of this thesis explores these misalignments through the development o
 f data-driven tools\, resulting in three systems that expand the design po
 ssibilities in mobile augmented reality\, create novel mechanisms for stre
 amer-audience engagement on live streaming platforms\, and support video c
 reators in expressing musical preferences through a vibe-based creative as
 sistant. Taken together\, my work on these projects revealed that the cont
 ent creation process produces a wealth of valuable data\, yet this data is
  often leveraged ad hoc to power isolated features\, as opposed to being t
 reated as a foundational resource for designing creator-aligned systems.\n
 \nBuilding upon this realization\, I argue that the future of creativity t
 ools relies on the deliberate use of Creative Activity Traces as a primary
  design material. The term Creative Activity Traces refers to the process-
 related data generated by the creator during their practice\, including ev
 olving records of ideas\, intermediate artifacts\, and metadata yielded du
 ring creative work. These traces are often multi-modal\, distributed acros
 s tools\, and deeply personalized. To understand how the HCI community cur
 rently works with traces\, I conducted a systematic review of 133 creativi
 ty tools and developed a preliminary design framework for trace awareness.
  This review revealed that modern creativity tools either treat activity t
 races as atemporal raw material\, or utilize them only as needed as input 
 to a specific feature\, thereby limiting both the temporal scope and desig
 n opportunities possible with traces. Rather than continuing to use traces
  in this way\, my thesis instead calls for traces to be treated as a desig
 n material to be engaged with at every stage of the research and developme
 nt process. This approach in turn leads to adaptive systems that stay alig
 ned with how creators work and how and their processes  unfold\,\n\nThe fi
 nal phase of this research builds upon my theory of creative activity trac
 es and trace-aware tools through the study of video content creators' prac
 tices for creating\, maintaining\, and evolving a recognizable brand acros
 s video projects. Through a formative study with seven video creators\, I 
 cataloged the traces their practice accumulates over time\, the behaviors 
 associated with their accumulated traces across projects\, and the frictio
 ns they face as a result of their cross-project practices. This work showe
 d that traces share temporal characteristics but are not a uniform materia
 l a designer can treat the same way throughout\, so much as a single mater
 ial with differing grains and textures. As traces accumulate across projec
 ts\, they weave into a whole with a shape of its own\, and the individual 
 traces that produced it can be difficult to disentangle into constituent p
 arts.\n\nThis thesis contributes to HCI and creativity tool research by fr
 aming creative practice as an inherently data-rich process that requires m
 ore intentional engagement from the HCI research community. I formalize th
 e definitions of creative activity traces and trace-aware tools\, and I of
 fer a design framework for trace-awareness. I show what it means to treat 
 traces as a critical building block for designing creativity tools rather 
 than as a byproduct of creative work. Taken together\, these contributions
  set forth a trace-aware agenda for the next generation of creativity tool
 s\, one that keeps creators and their evolving practices in view as digita
 l creative production grows more automated and uncertain.\n\nThesis Commit
